SFARI iPSC Resource

Tab 1 lists all iPSCs that are created at the New York Stem Cell Foundation. This effort is co-funded with the Nancy Lurie Marks Family Foundation (NLMFF).
- At the NYSCF PBMCs are reprogrammed using Sendai Virus deliver of reprogramming factors. The repogramming process takes 6-9 months, including in depth Quality Control (QC). If no clone is found to meet QC criteria a new clone is run through the QC pipeline, which adds an additional 3 months.
- Genetic variants to create iPSCs for, are chosen based on blood sample availability, active participation in Simons Searchlight, SFARI scientific priorities, researcher interest and family group leader feedback
- if a large number of blood samples is available for one gene, we use the following prioritization criteria for variants: de novo predicted loss of function (early STOP, splice site variants) > de novo recurrent missense (predicted to disrupt protein function) > de novo non recurrent missense (predicted to disrupt protein function) > de novo non recurrent missense (VUS) > inherited variants

Tabs 3, 4 and 5 list additional iPSC lines that are available through SFARI Base but not created through the collaboration with the NYSCF. Those tabs also list iPSC lines created from participants in the Simons Simplex Collection. Note that for some of the genes represented on Tab 1, we have additional iPSCs listed on Tab 3

iPSCs are stored and distributed at/by 'Sampled' and can be requested by any qualified researcher through SFARI Base. If a line/clone is requestable is indicated on each tab in a column labeled "available for request on SFARI Base". Clones are only available for request if they passed all QC.

iPSCs lines from sex matched family members can be identified using the SFARI-IDs. The first five digits of the SFARI ID are identical for family members.
