1. DNA nanoballs are generated by linear rolling circle amplification (RCA). RCA avoids the exponential accumulation of errors, GC biases observed with other amplification methods, such as PCR.
2. Only one strand of the dsDNA is used for circularization as the subsequent sequencing template. This results in a significant reduction in duplication rate and more effective usage in data. As a result, it enables generation of more data with lower cost on MGI platform.
3. Without accumulation of PCR errors, index hopping rate can be reduced to 0.0001%-0.0004% using single index.
Using bridge PCR to generate DNA clusters on flow cell.
1. bridge PCR amplification can produce exponential accumulation of errors.
2. Two identical clusters can be generated by the complementary strands of dsDNA library, increasing in duplication rate.
3. Using Examp technology, index hopping rate can reach 1%-6% with single index.

Flow cell TechnologyPatterned array
Optimized patterned array ensures that only one single DNB is attached at each spot, which results in better flow cell utilization with unprecedented uniformity. This enables a significant reduction in duplicate rate.
