What to do
2
Make a copy of this sheet
3
If you are working with individual samples, select the "25B-Ind-ReadsPerLane" or "5B-Ind-ReadsPerLane". If you are working with PoolSeq samples, select "25B-PoolSeq" and "5B-PoolSeq" tabs. Each tab differs in the number of reads per lane. The goal is to calculate the cheapest option that gives the minimum optimal coverage.
4
In each of the "25B" tab and the "5B" tabs, enter the number of samples, and the genome size
5
Check that the amounts in the green boxes will work. Please make sure you have at least 12x coverage for individual samples (15x is better) and 100x coverage per pool for pooled samples
6
Choose the tab with the lowest total price for the project. The tabs differ in the sequencing costs, with lower costs for deeper sequencing. But for smaller genomes, sequencing at that depth may not be needed.
7
Only adjust the QTY in orange if you are requesting more than $5.2K for a sequencing grant, or if your genome size is so small the total is less tha $5.2K
8
Before you submit, delete the tabs that you're not using. For example, if the tab "25B" has a lower cost, delete the tab "5B".
9
Don't forget to enter your contact info and the MacroMarID in the tab
