Sequencing principleUses clonal bridge amplication to form DNA clusters on flow cells with pre-attached short oligos
1. Prone to PCR bias and error accumulation.
2. Increased duplication reads, arising from optical duplicates (a common problem associated with randomly clustered flow cells), and complementary strands of the same library generating independent clusters.
3. Miseq single index misassignment rates range from 0.1-0.5%. ExAmp single index misassignment rates range from 1-6%. (Note that ExAmp is only used on HiSeq 3000/4000/X)
Uses DNBSEQ™ technology - DNA Nanoballs (DNB) are formed by rolling circle amplification, and adhered to a patterned array flow cell for sequencing.
1. Amplification is based on original template. This results in linear amplification with low amplification bias and low error accumulation.
2. Low duplication rates due to patterned array configuration. Also, only one strand (the sense strand) is used for circularisation, hence duplication rates on MGI is far lower than other platforms, increasing the read efficiency, and reducing data wastage.
3. Without accumulation of PCR errors, index hopping rate can be reduced to 0.0001% to 0.0004% using a single index.
