H7 is potential candidate for scaffolding for universal vaccine design

  • General human population is not exposed to H7 protein

  • H7 elicits cross-group stem specific immune response

  • Influenza H7N9 epidemic is gaining momentum

Background

  • HA has head and stem domains
  • Head: strain-specific, immunodominant
  • Stem: highly conserved, immunosubdominant
  • We can shield head epitopes from antibodies using engineered glycans

Outline

  1. Test the TrimerTag for H7 expression
  2. Use the TrimerTag to express 7 different Has
  3. Characterize the H7 head- and stem-only proteins

Part 1: TrimerTag

  • Current trimerization strategies in the lab:
    • HA-foldon
    • HA-IZ
    • HA-Ferritin
  • In this project, we wanted to test a new trimerization domain, the TrimerTag

Using Biotin Anti-C-tag Conjugate to load C-tagged proteins

onto Octet Streptavidin Tips

CaptureSelect™ Biotin Anti-C-tag Conjugate consists of a 13 kDa camelid antibody fragment (affinity ligand) with high affinity and selectivity for "C-tag" peptide tag E‑P‑E‑A (glutamic acid - proline - glutamic acid - alanine).

Part 1 Conclusion

  • H7-TrimerTag binds to well-characterized stem and head antibodies
  • The current H7+10 constructs have unprotected head epitopes
    • Steven is testing new H7+X constructs
  • The TrimerTag can be used as an alternative trimerization strategy for HA expression

Part 2: Testing 7 different HAs

  • Test whether well-characterized antibodies against H7 demonstrate expected cross-reactivity against other HAs

Part 2 Conclusions

  • The TrimerTag can be used to express HAs from diverse strains of influenza
  • The purified HAs bind to well-characterized bNAbs
  • The purified HAs will be used as a substrate for testing sera binding from H7-immunized mice

Part 3 Conclusions

  • The purified head proteins showed specificity to head antibodies compared to the full length H7 foldon, which bound to both head and stem antibodies
    • Use the purified samples in high-throughput screens to profile antibodies.
