iGEM BioDesign Bootcamp Application 2019
The iGEM BioDesign Bootcamp is an intensive introduction to synthetic biology, an emerging field which uses molecular techniques to engineer biological systems that solve problems. This Wintersession program, which runs from January 19th to January 25th, will enable participants to learn foundation research skills and get their hands wet with genetic engineering. In parallel to a laboratory component which will guide students through the construction of a genetic device, Professor Neel Joshi of the Wyss Institute and SEAS will provide a conceptual introduction to the field and guide students through the process of project design. Participants will learn widely applicable laboratory skills and protocols which make up the "toolbox" of synthetic biology, including PCR (polymerase chain reaction), DNA purification, bacterial culture and cloning. Although the tempo will be fast-paced, this experience is geared towards participants with diverse backgrounds and any level of prior experience with biological research, including none at all. Resources will be provided to get everyone up to speed.

This program is sponsored by Harvard iGEM and will be excellent preparation for students interested in competing on the 2019 Harvard iGEM team. If students desire future research opportunities, they will be put in contact with the iGEM club, undergraduate research coursework like LS100-r, and faculty laboratories.

Given the following scenario, figure out why the experiment didn't work. What did the iGEMers do wrong?
The iGEM team 2018 is in the iGEM lab working on creating the next big thing! The team is setting up to do transformations which would make their bacteria Kanamycin resistant. Kanamycin is an antibiotic that would kill bacteria. The gene for kanamycin resistance is often abbreviated as Kan. They grab a tube labeled 'Kan' and leave it to defrost on a bucket of ice. They grab a culture of bacteria suspended and checked the density of it. You notice that it seems really bright in the room today. You read the sample and it says that the density is at 0.8. Yay! You go on to the next part of the experiment. You accidentally left the sample to defrost an extra minute. You go on with making the bacteria antibiotic resistant anyways. You pipette the bacteria and the Kan into a tube and shock it. You immediately put broth into your cell and place it at the right temperature to make them grow . You plate them for overnight incubation in a kanymycin plate. The next day you come back, and you are sad to find that no colonies grew. Despite your experiment failing, you are determined to try again!
