1. Place the strain in LB for bacteria and Knockout media for yeast or other appropriate media
  2. Add the appropriate antibiotic for bacteria in the correct concentration, this can be found by searching the internet for “<antibiotic name> working concentration” where <antibiotic name> is the name of the antibiotic need for plasmid selection
  3. If bacteria or yeast don’t grow or take a long time to grow, first create a starter culture by inoculating media without an antibiotic for 1-2 hours and allowing the strain to grow then adding 100uL / mL to the selection media.
